So, the default UI places your target way the heck up in the corner, and your ability buttons (where all the cooldown timers you need to watch are) down at the bottom...

Target on the right of my ability bars. Target's target on the left of my ability bars, focus below that.
Abilities set up to be easy to hit with chording. I have a colemak keyboard, so my layout near my left hand is:

arst

WARS to move. F for spam skills (combo builders on rogue) Q for finishers, P & T for useful skills in my rotation. ` for "Oh crap" buttons, 1, 2, & 3 for things like interrupts and DPS cooldowns I probably don't need.
All can be modified by Shift, Ctrl, Alt, or any pair of those. So for each button I get 7 buttons.
Right side bars are utility stuff, bound to the numpad + modifiers.
Shift + Movement keys (WARS) are also bound to functions. Shift + S for a potion, for example.
My left hand stays still, my mouse is free to move, I can run around and hit abilities at the same time, and everything I need to see is easily visible.

I have my frame and target from in the default positions, but I put my minimap in bottom center with two ability bars below it. Then I have ability bars on either side of those (so 6 total on bottom) with a chat box on either side (one for general chat stuff, one for guilds, raid, party, etc). Also, this site has a bunch of different UI setups that can be loaded (or to get ideas from): http://riftlift.com/
